MicroRNAs are implicated in the response to biotic stresses. Papaya meleira virus (PMeV) is the causal agent of sticky disease, a commercially important pathology in papaya for which there are currently no resistant varieties. PMeV has a number of unusual features, such as residence in the laticifers of infected plants, and the response of the papaya to PMeV infection is not well understood. The protein levels of 20S proteasome subunits increase during PMeV infection, suggesting that proteolysis could be an important aspect of the plant defense response mechanism. To date, 10,598 plant microRNAs have been identified in the Plant miRNAs Database, but only two, miR162 and miR403, are from papaya. In this study, known plant microRNA sequences were used to search for potential microRNAs in the papaya genome. A total of 462 microRNAs, representing 72 microRNA families, were identified. The expression of 11 microRNAs, whose targets are involved in 20S and 26S proteasomal degradation and in other stress response pathways, was compared by real-time PCR in healthy and infected papaya leaf tissue. We found that the expression of miRNAs involved in proteasomal degradation increased in response to very low levels of PMeV titre and decreased as the viral titre increased. In contrast, miRNAs implicated in the plant response to biotic stress decreased their expression at very low level of PMeV and increased at high PMeV levels. Corroborating with this results, analysed target genes for this miRNAs had their expression modulated in a dependent manner. This study represents a comprehensive identification of conserved miRNAs inpapaya. The data presented here might help to complement the available molecular and genomic tools for the study of papaya. The differential expression of some miRNAs and identifying their target genes will be helpful for understanding the regulation and interaction of PMeV and papaya. O Espírito Santo se destaca como o maior produtor brasileiro da espécie Coffea canephora, conhecida no Estado como ?Conilon?. Este trabalho objetivou avaliar a performance e estimar ganhos de seleção de uma população de maturação tardia de Coffea canephora, variedade Conilon, no primeiro ciclo de seleção recorrente oriunda do programa de melhoramento genético do Instituto Capixaba de Pesquisa, Assistência Técnica e Extensão Rural (Incaper). Foram avaliadas 1.019 progênies de 11 famílias de meios-irmãos provenientes da recombinação em campo isolado, de 11 progenitores promissores do programa de melhoramento genético, com a utilização da estratégia de seleção recorrente intrapopulacional. Observou-se comportamento médio das progênies recombinadas (PR) superior aos dos progenitores (P) para reação à ferrugem, produção de grãos por planta, tamanho e uniformidade de maturação dos frutos, evidenciando menor incidência de ferrugem nas progênies recombinadas, maior produção, maturação mais tardia e uniforme. A intensidade de seleção que determinou maior ganho genético foi a de 45% entre e 20% dentro de famílias de meios-irmãos, com ganho de 10,46% para produção de grãos.
The Espirito Santo stands out as the major producer of Coffea canephora species, known in the state as 'Conilon'. This study evaluated the performance and estimate selection gain to late ripening population of Coffea canephora, in the first cycle of recurrent selection, coming from the breeding program of the Capixaba Research Institute, Technical Assistance and Rural Extension (INCAPER). 1.019 progenies of 11 families of half-sib were evaluated from recombination in isolated field, of 11 promising breeding program parents, using the intrapopulation recurrent strategy selection. The average behavior of recombinant progeny (PR) showed better results than the parent (P) for grain yield per plant, size and uniformity of fruits ripening and reaction to rust, showing greater production, late maturation, more uniform maturation and lower incidence of rust in recombinant progeny. The intensity selection that determined higher genetic gain was 45% between and 20% within half-sib families, with a gain of 10.46% for grain production.
